CILOSITAZONE is a medication primarily used to treat intermittent claudication, a condition characterized by pain, cramps, numbness, or weakness in the legs due to poor blood circulation, typically caused by peripheral artery disease (PAD). CILOSITAZONE works by inhibiting phosphodiesterase type 3 (PDE3). This inhibition leads to an increase in cyclic adenosine monophosphate (cAMP) within vascular smooth muscle cells and platelets, which, in turn, has several pharmacological effects:
Mechanism of Action
Vasodilation: CILOSITAZONE causes the blood vessels to widen, improving blood flow to the limbs.
Inhibition of Platelet Aggregation: It reduces the ability of platelets to stick together, decreasing the risk of blood clots forming in the arteries.
Increased Walking Distance: By improving blood flow and reducing blood clotting, cilostazol helps people with intermittent claudication walk longer distances without pain.
Clinical Use
CILOSITAZONE is specifically indicated for the improvement of the symptoms of intermittent claudication as evidenced by an increased walking distance. It is not recommended for patients with heart failure of any severity due to the risk of increased mortality with PDE3 inhibitors in this patient population. The treatment with cilostazol is part of a comprehensive management plan for PAD, which also includes lifestyle changes like smoking cessation, exercise, and diet modifications.
